Abstract
Safranin is a kind of synthetic dye that is commonly used in gram staining. However, with rising prices and difficulties in obtaining these synthetic dyes in remote areas, alternatives to synthetic dyes are needed. One reliable solution is to use natural dyes that come from natural sources. Natural ingredients that can be used as a substitute for synthetic dyes are anthocyanins found in the skin of the mangosteen fruit (Garcinia mangostana L). Mangosteen contains about 593 ppm of anthocyanins, which are natural pigment compounds with coloring abilities. Through the method of extracting anthocyanins from mangosteen rind, natural dyes in the form of anthocyanin pigments can be produced. These pigments are capable of giving red, purple, and blue colors, and can be used as a gram stain for bacteria. The use of anthocyanins from mangosteen rind as natural dyes offers several advantages, such as easy-to-obtain and environmentally friendly sources. Thus, the use of natural dyes can be a good solution to overcome the difficulty of obtaining synthetic dyes and help reduce dependence on synthetic-based chemicals. This study aims to evaluate the ability of the results of mangosteen peel extraction as an alternative dye for gram staining, as a substitute for safranin, against Staphylococcus aureus and Escherichia coli bacteria. The research method involved extracting macerated mangosteen rind, then diluting it in various concentrations, namely 10%, 30%, 50%, 70%, and 100%. As a positive control, synthetic dyes were used. Data were in the form of tables and descriptions of paintings with good, bad, and bad criteria. The results obtained at concentrations of 10% and 30% showed that mangosteen rind instead of safranin dye was able to color bacteria but was less effective as a substitute dye because the absorbed color lacked contrast.
