Abstract

Candida albicans is a member of the normal flora found in mucous membranes, digestive tract, respiratory tract, urethra, vagina, skin and fingernails and toenails. Candida albicans can grow at a wide range of pH, but generally growth will be better at a pH between 4,5-6,5. Candida albicans can grow in seed at a temperature of 28ºC – 37ºC. The type of research used is descriptive quantitative research. This study aims to determine the presence of the fungus Candida albicans in the urine of patients with urinary tract infections using the Real Time PCR method. The population in the study was patients with urinary tract infections. Urinary tract infection is an infection with clinical conditions due to the proliferation of microorganisms that cause inflammation in the urinary tract. The research was carried out at the Parasitology Laboratory and Molecular Biology Laboratory from April 2022 to May 2022. In this study, several stages of examination were carried out such as isolation of culture on Sabouroud Dextrose Agar media, macroscopic and microscopic examinations, manufacture of cell suspensions, extraction tests, purity tests DNA, optimization and Real Time PCR examination by showing results in the form of CT. The results showed that during the Real Time PCR examination, 2 samples with a percentage of 22,2% were obtained in the sample code S7 with a CT value of 28,50 and the sample code S9 with a CT value of 23,19 and a negative result of 7 samples with a percentage of 77,8 %. The causative factors that can lead to the presence of the fungus Candida albicans in the urine of patients with urinary tract infections such as personal hygiene, people with diabetes mellitus, contraceptives and the use of antiseptics.
